Description

Principle of the Assay::The Human Neuregulin-1 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human Neuregulin-1 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human Neuregulin-1 coated on a 96-well plate. Standards and samples are pipetted into the wells and Neuregulin-1 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human Neuregulin-1 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of Neuregulin-1 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Neuregulin 1 or NRG1 is a protein that in humans is encoded by the NRG1 gene. It is mapped to 8p12. NRG1 is one of four proteins in the neuregulin family that act on the EGFR family of receptors. The protein encoded by this gene is a membrane glycoprotein that mediates cell-cell signaling and plays a critical role in the growth and development of multiple organ systems. Dysregulation of this gene has been linked to diseases such as cancer, schizophrenia, and bipolar disorder (BPD). It is also essential for the normal development of the nervous system and the heart. Furthermore, it was originally identified as a 44-kD glycoprotein that interacts with the NEU/ERBB2 receptor tyrosine kinase to increase its phosphorylation on tyrosine residues
