Principle of the Assay::The Mouse CD144 ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse CD144 in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Mouse CD144 coated on a 96-well plate. Standards and samples are pipetted into the wells and CD144 present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse CD144 antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of CD144 bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.

Background/Introduction: Cadherin 5, type 2 or VE-cadherin also known as CD144, is encoded by the gene CDH5. It is a classical cadherin from the cadherin superfamily. This gene is mapped to mouse chromosome 8. The encoded protein is a calcium-dependent cell-cell adhesion glycoprotein comprised of five extracellular cadherin repeats, a transmembrane region and a highly conserved cytoplasmic tail. Functioning as a classic cadherin by imparting to cells the ability to adhere in a homophilic manner, the protein may play an important role in endothelial cell biology through control of theorganization of the intercellular junctions. It has found that PECAM1, VE-cadherin and VEGFR2 together are sufficient to confer responsiveness to flow in heterologous cells
