Principle of the Assay::The Human MBL2 Binding Protein ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Human MBL2 Binding Protein in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Human MBL2 Binding Protein coated on a 96-well plate. Standards and samples are pipetted into the wells and MBL2 Binding Protein present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Human MBL2 Binding Protein antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of MBL2 Binding Protein bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Background/Introduction: MBL2, also called mannose-binding lectin (protein C) 2, soluble or Mannose-binding lectin (MBL) is a lectin that is instrumental in innate immunity. MBL2 is mapped to chromosome 10q11.2-q21. It belongs to the class of collectins in the C-type lectin superfamily, whose function appears to be pattern recognition in the first line of defense in the pre-immune host. MBL2 recognizes carbohydrate patterns, found on the surface of a large number of pathogenic micro-organisms, including bacteria, viruses, protozoa and fungi. Binding MBL2 to a micro-organism results in activation of the lectin pathway of the complement system. Another important function of MBL2 is that this molecule binds senescent and apoptotic cells and enhances engulfment of whole, intact apoptotic cells, as well as cell debris by phagocytes
