Principle of the Assay::The Mouse IL-17C ELISA (Enzyme-Linked Immunosorbent Assay) kit is an in vitro enzyme-linked immunosorbent assay for the quantitative measurement of Mouse IL-17C in Cell Culture Supernatants, Serum, Plasma. This assay employs an antibody specific for Mouse IL-17C coated on a 96-well plate. Standards and samples are pipetted into the wells and IL-17C present in a sample is bound to the wells by the immobilized antibody. The wells are washed and biotinylated anti-Mouse IL-17C antibody is added. After washing away unbound biotinylated antibody, HRP-conjugated streptavidin is pipetted to the wells. The wells are again washed, a TMB substrate solution is added to the wells and color develops in proportion to the amount of IL-17C bound. The Stop Solution changes the color from blue to yellow, and the intensity of the color is measured at 450 nm.
Background/Introduction: IL17C, also known as CX2, is a protein that in humans is encoded by the IL17C gene. IL17C is mapped to 16q24.3. The protein encoded by this gene is a T cell-derived cytokine that shares the sequence similarity with IL17. This cytokine was reported to stimulate the release of tumor necrosis factor alpha and interleukin 1 beta from a monocytic cell line. The expression of this cytokine was found to be restricted to activated T cells. IL17C is an essential autocrine cytokine regulating innate epithelial immune responses. It also plays an important role in the pathogenesis of inflammatory arthritis
