Mouse Factor IX Genetically Deficient Brain Tissue Lysate is prepared from a Factor IX knockout mouse. This Factor IX Knockout was created by targeted disruption of the F9 gene. Mutations were then confirmed by PCR genotyping, Sanger sequencing, and RT-PCR. Organ is flash-frozen immediately upon removal and stored frozen until lysed. The tissue is then lyophilized and thorougly homogenized in Radio Immuno Precipitation Assay (RIPA) buffer containing protease and phosphate incubators. The final homogenate is then incubated with DNase, clarified via centrifugation (10,000g at 4 degree C) and stored at-80 degree C. Each lot is tested for protein concentration via BCA assay and analyzed via SDS-PAGE to confirm the presence of sharp, clear bands as an indicator of quality
