Description

Automated acquisition, analysis, and reporting for hands-free flow cytometry

Express Modes are a unique add-on for the MACSQuantify™ Software, developed to simplify flow cytometric analysis. They automate the measurement and analysis of flow experiments via predefined experiment settings and acquisition and automated analysis. A gating to your results will be automatically adjusted for each data file individually to achieve optimal results. Using the Express Modes simplifies flow cytometric analysis and ensures the reproducible data analysis.

Viral or fungal infections can lead to major complications in the period of immune recovery after hematopoietic stem cell transplantation (HSCT). Adoptively transferred antigen-specific T cells (CCS) have been shown to restore protective immunity and control established adenovirus (AdV), cytomegalovirus (CMV), and Epstein-Barr virus (EBV) infections after HSCT. Virus-specific T cells can be enriched based on their secretion of IFN-gamma after restimulation with the appropriate antigen by using the CliniMACS® Cytokine Capture System (IFN-gamma) (CCS). This technology easily allows the enrichment of virus-specific CD4 and CD8 T cells of various specificities. To determine the success of the enrichment, different cell fractions need to be analyzed with regard to the cellular composition, the target cell number, and the purity.

For standardization and automation of this analysis, Virus-Specific T Cell CCS Express Modes were developed.
