IL1 receptor antagonist (IL1RN) is a protein that binds to IL1 receptors (IL1R1) and inhibits the binding of IL1-alpha and IL1-beta (IL1B). It specifically inhibited IL1 bioactivity on T cells and endothelial cells in vitro and was a potent inhibitor of IL1-induced corticosterone production in vivo. High concentrations of glucose induce beta cell production of IL-1beta, leading to impaired beta cell function and apoptosis in human pancreatic islets. IL-1 receptor antagonist (IL-1Ra) is a naturally occurring antagonist of IL-1beta and protects cultured human islets from glucotoxicity. Therefore, the balance of IL-1beta and IL-1Ra may play a crucial role in the pathogenesis of diabetes.
