The canonical protein structure is reported to have an amino acid length of 1106 residues, a mass of 117.9 kDa, and is a member of the GLI C2H2-type zinc-finger protein family. It is known to be localized in the nucleus and cytoplasm and is notably expressed in testis. As many as 3 protein isoforms have been reported. The protein has been noted to be post-translationally ubiquitinated, acetylated and phosphorylated. The GLI1 gene has been associated with the disease, Polydactyly.
