We have been using this ACK lysing buffer for several projects. Whenever we need to isolate single cells from mouse organs, we will use this ACK lysing buffer to lysis red blood cells. It worked very well.
Lysis of red blood cells
RBCs
Repeat with lysis buffer if red blood cells persist after initial lysis.
We can effectively remove red blood cells by mixing this buffer with the isolated cells from organs. The remaining cells are unaffected and still maintain good viability.
