We are trying to determine the 50% effective concentration value (EC50) of a certain compound against Dengue virus by immunofluorescence assay. We used this primary antibody from Novus, it is conjugated to Alexa Fluor-488 and it targets the envelope protein of flavivirus. During our assay optimization, we determine optimal viral dilution correspondent to 15% of infected cells. We use 1:1000 dilution and it works well and gives a reliable signal.

This assay allowed us to determine 50% effective concentration (EC50) of an antiviral compound against Dengue virus. We added different compound concentrations to virus and correlated fluorescence signal to compound concentration, which generated a dose-response curve and corresponding EC50 value.
