Product Name:

ACK Lysing Buffer, 500mL

Catalog Number:

118-156-101

I used this buffer to lyse the red blood cells that were in my digested tumor tissue. Depending on the size of the tumor, you may need to repeat lysis but I found that the product works well without causing a significant drop in final cell viability.

Experimental Design and Results Summary

Application

Lysing of red blood cells

Starting Material

Tumors and spleen from mice

Tips

Do not extend lysis for more than 5 minutes.

Results Summary

This product lysed the red blood cells in my samples and create single cell suspensions, which I could then stain for and run analysis by flow cytometry.

Summary

The Good

Affordable and can be stored at RT.

The Bad

May require repeated lysis.

The Bottom Line

This product works to generate single cell suspension from digested or processed tissues. While it works, some samples may require a second lysis step so you'll need to be mindful of that while also making sure not to over lyse your sample. In general, this is an efficient ACK lysis buffer I would recommend to others.
