This kit is used daily in my lab, it allows purification of PCR products in a fast and easy way. I follow the supplier's protocol with the following modifications: - After diluting the PCR reaction with buffer PB, I pass this mixture twice through the column. This allows more DNA to bind to the silica. - After washing with buffer PE, usually, there is still some buffer (~2 uL) around the silica that needs to be removed, just use a pipette for that. - After washing, let the silica air dry to allow ethanol evaporation before moving to elution. - Pre-warm your elution buffer or water to 60 °C before adding to the column and let it sit there for around 3-5 min. It helps yield more DNA. - I always elute my DNA twice, it helps get more DNA out of the silica. -The protocol recommends elution in 50 uL, I usually elute with 35 uL, it can help increase concentration if that is your goal. I wouldn't use less than 35 uL because the silica would not be wet enough. The picture represents the DNA concentration I got from a 100 uL PCR reaction, following the steps I described above. This kit always gives consistent results and good DNA quality.
