I have examined the physiological roles of two E. coli proteins with unknown functions, MepK and YgeR, in cell wall metabolism. Initially, I assessed their overexpression, and solubility, and then carried out protein purification. Subsequently, I analyzed their biochemical activities on purified bacterial cell walls. To assess overexpression and solubility, and during various chromatography procedures, I utilized a premixed 4x Laemmli buffer to prepare samples for SDS-PAGE.

I have discovered that the protein with peptidoglycan (PG) hydrolytic activity, MepK, displays a substrate specificity for 3-3 cross-linked muropeptides (L, D-endopeptidase). It plays a role in the expansion of the PG sacculus, working in conjunction with other cell wall hydroalses (D,D-endopeptidases). This underscores the essential significance of cross-link cleavage in bacterial PG synthesis.
