I have biochemically characterized a cell wall hydrolase, MepK. To assess its in vitro activity, I initially performed a Zymogram analysis to examine the functionality of the purified enzyme. Zymogram analysis is a widely used technique in bacterial cell wall research, particularly for studying enzymes like cell wall hydrolases. These enzymes cleave the bacterial cell wall. Zymogram analysis enables researchers to identify and comprehend the activity of these enzymes by observing the zone of clearance on the gel where the substrate (cell wall material or Micrococcus lysodeikticus cells) is digested due to the enzyme's effect.

Gently sonicate the Micrococcus lysodeikticus cells and thoroughly mix the cell lysate with the SDS-polyacrylamide gels before casting them.

Results Summary

In this gel-based assay, MepK created a zone of clearance in the gel that had Micrococcus lysodeikticus cell lysate and was stained with methylene blue. Like lysozyme and MepM (positive controls), purified MepK also displayed a clearance area, suggesting it might have cell wall-breaking abilities. BSA served as the negative control.
