Spin down supernatant prior to use, and aliquot in small volumes. Thaw supernatants only once.

Results Summary

I found that HMGB1 release was higher at higher temperatures of heating, with a drop off at 85 degrees C. I also found that irradiation of these radioresistant cells produced higher HMGB1 release with low irradiation dose, but could not interpret a dose-dependent HMGB1 release with higher irradiation doses.

Higher concentrations detected than other kits, also didn't require dilution.

The Bad

Takes 3 days.

The Bottom Line

If you're willing to take 3 days on an ELISA, this isn't a bad kit. However, there are other kits on the market that come pre-coated or are otherwise optimized to take less time. This kit is easy to use and simple, and performs fairly well, but it doesn't seem to be any better than other kits with a similar range of detection.
