I use precise genome editing to generate endogenous mutations in the components of the WNT pathway. My previous attempts using the CRISPR/Cas9 RNP format yielded poor knock-in efficiencies. However, my success rate got better by including the IDT HDR Enhancer V2 reagent.

Results Summary

500,000 HAP1 cells were electroporated with 100 pool of CRISPR/Cas9 RNP and 1 µM of IDT HDR Enhancer V2. Medium was changed after 24 hours but the reagent was included for 48 more hours. 72 hours post electroporation, INDEL and Knock-In efficiencies were determined from Sanger Sequencing traces.
