A great KIT for imaging end-stage apoptosis in tissues or cells (TUNEL technique). It gives very good results in tissues that have been fixed in formol and blocked in paraffin. The most important point is the antigen detection technique applied in the tissues with formol fixation. Otherwise, intense nonspecific nuclear staining is obtained.

Results Summary

A great KIT for demonstrating end-stage apoptosis, a method that works on any tissue type. I use it frequently in my lab. Ideal for staining 50+5 medium sized tissues together with the control. If you shake off the liquid remaining in the tissue during the washing stages, you can stain +10 extra tissue. It should be noted that Proteinase K administration should be nuclease free and adjusted at a dose of 10 ug/ml. This application is sufficient for 10 minutes at room temperature. However, it should not be forgotten that various laboratory conditions such as long formol fixation time, quality and dilution of formol can change these times. Each laboratory should set its own. However, excessive digestion causes severe nuclear nonspecific staining. This step should be given great attention.

Summary

The Good

Very specific signaling for end-stage apoptosis.

The Bad

Difficult to optimize.

The Bottom Line

It is a very expensive kit, the amount of testing is small, but when the technique is applied well, it is very effective in detecting end-stage apoptosis.
