We are an immunotherapy focused group, with interest in CD8 T-cells and their TCRs. For our experimental purposes, we routinely transduce TCRs in PMBCs, with the viral particles being produced by AmphoPack-293s transfected with the relevant plasmids.

Use antibiotics-free media
5million AmphoPack-293 were transfected with the relevant plasmids, plus 0,2ug GFP. We commonly see >80% gfp positive cells after 2 days of culturing.

This reagent works well for plasmid transfection. Note that not all cell lines will be easy to transfect with such polymer-based reagents.
