Company:

Biotium

Product Name:

MemBrite Fix 594/615

Catalog Number:

30096-T

Primary T cells were stained with MemBrite Fix Cell Surface Stain and cultured on unstained primary endothelial cells to analyze T cell lateral and transendothelial migration by live fluorescent microscopy.

Experimental Design and Results Summary

Application

Fluorescent microscopy

Starting Material

Primary mouse T cells

Tips

Stain becomes internalized by cells quickly.

Results Summary

MemBrite stains cells brightly and does not appear to affect membrane ruffling and migration of T cells. Surface stain becomes internalized by T cells within 30 minutes of staining and appears as bright punctae inside cells instead of surface stain.

Summary

The Good

Quick staining that does not require pretreatment of cells.

The Bad

Surface stain is internalized by cells rapidly.

The Bottom Line

MemBrite Fix stain works very well to label cells for tracking movement and migration by live fluorescent microscopy. However, the surface stain gets internalized by some cells rapidly and appears as cytoplasmic punctae.
