The work of our group is largely based on vectors and plasmids we use for transfections in human cell lines. To prepare our plasmids from Bacteria we use the NucleoBond Xtra Midi Plus Kit and are in general quite satified with the results.
Plasmid preparation
Bacterial culture transformed with plasmid of interest.
We always use the volumes for Midiprep (not Maxiprep) and start with a 100 - 200 ml Bacteria culture grown for approx 24 h.
After Midiprep we obtained clean plasmids in decent but reliable concentrations (yield could be better but you might just use more bacteria as starting material).

Easy protocol, almost no additional equipment needed.
Price, a lot of material (waste).
The Kit basically does the job without much optimization and equipment needed and plasmids are clean and ready to use.
