We enriched Natural Killer cells from mouse splenocytes to use for a future functional assay. We compared the percentage of Natural Killer cells that made up a sample of lymphocytes before and after using the antibody cocktail in the mouse NK cell isolation kit.

Used 10 µl of antibody cocktail and beads for 1 X 10^6 cells in 100 µl of buffer

Results Summary

We saw a significantly better percentage of Natural killer cells in the splenocytes after using the NK cell isolation kit. We saw 47% of NK cells prior to enrichment and 86% after enrichment. This allowed for more accurate results in the proceeding functional assay.

Summary

The Good

For the most part the percentage of enriched NK cell was around 80%.

The Bad

We lost some of our NK cells that are selected through antibodies in the kit cocktail.

The Bottom Line

The NK antibody cocktail works great, although I think less antibody than what is recommended in the protocol could be used to achieve similar results.
