We checked the expression of LGALS-9 (GAL-9) on non-parenchymal mouse liver cells. Gal-9 is a checkpoint inhibitor of CD8+ T cells. The aim of our experiment was to identify which cell types predominantly express GAL-9 in liver.

Experimental Design and Results Summary

Applications

Flow Cytometry

Sample

Mus musculus C57/BL6, Non-parenchymal liver cells

Primary Incubation

1:100 dilution, 30 minutes at 4 degree Celsius

Blocking Agent

FcR blocking reagent

Secondary Incubation

Not needed

Tertiary Incubation

Not needed

Detection

Flow cytometry detection by Per-Cp channel

Results Summary

Non-parenchymal liver cells were analysed for surface expression of Gal-9 by flow cytometry. We compared the Liver Sinucidal endothelial cells (LSECs) and Kupffer Cells (KCs) for the surface expression of same. Our results indicated that KCs express Gal-9 predominantly than LSECs in mouse liver. The figures show positive population as compared to the negative population. An elevated expression of Gal-9 on surface indicate possible suppression of CD8+ T cell response by checkpoint inhibition.

Summary

The Good

Easy to use, less incubation time, less spillover , reliable

The Bad

moderate quality of results, quite expensive

The Bottom Line

We have noted synonymous results upon repeating, making this product reliable. It is worth to spend this much amount for an important or a confirmatory experiment. Also, the antibody works below recommended concentration but FMOs are necessary.
