I am studying a protein that is usually responsible for male gonad differentiation in mammals but is recently found out to be implicated in Parkinson's disease (PD). This protein is not present in the flies and I am trying to express it in the brain of the animal to check if its expression would cause changes/phenotypes that can be linked to PD.

Experimental Design and Results Summary

Application

Check presence of absence of protein bands

Starting Material

Immunoprecipitated protein

Tips

2-3 mins develop time (as indicated in protocol) is usually not enough for bands to be visible, give it at least 10 mins but be careful not to overdevelop it as it darkens so fast once the bands start to appear.

Results Summary

Presence of the protein was detected at the 50kDa mark

Summary

The Good

Easy to use, not messy (unlike in Coomassie Blue)

The Bad

Must be careful in timing the developing of the gel

The Bottom Line

Easy to use staining kit for protein detection but need to be wary of the time you develop your gels. The 2-3 min time is not enough for the bands to appear. It may take a longer time but when the bands start to appear be careful not to overdevelop them.
