We had great success using the Tyto to sort for Nuclei from multiple tissue types (both human and mouse). The Tyto is extremely user friendly and the sorts were quick (1ml/15 min). With the low pressure sorting cartridge system, the nuclei remained intact with little blebbing. Post-sort nuclei yields increased by 30-35%, allowing for us to perform robust single-cell assays.

1ml/15min is on the slower side, but Miltenyi addressed the issue with higher throughput cartridges

The Bottom Line

This system is perfect for sorting specific populations and since the sample sorts into a negative chamber, if any issue arises it is easy to re-sort the sample with no sample loss.
