Mouse glioma CT2A cells were treated with Sorafenib for 3 hours and its effect on phospho PI3K levels was examined. Equal amount of untreated and treated cells were subjected to Western blot analysis.
Western Blot
Untreated and sorafenib-treated CT2A cells
1:1000 dilution, 18 hours at 4 degrees with gentle shaking
5% non-fat dry milk in Tris buffer containing 0.1% Tween-20
HRP-conjugated anti-rabbit antibody 1:2000 dilution, 1 hour at room temperature with gentle shaking

ECL based detection
This antibody successfully detected the PI3 Kinase levels in CT2A cells. Low dosage of Sorafenib did not seem to have any effect on the levels of PI3 Kinase in CT2A cells.
High sensitivity
Some background and and non-specific bands were observed.
Its a great antibody for detection of PI3 Kinase p85 subunit in CT2A cells.
