Apoptosis/ Necrosis Assay Kit

Catalog Number:

This Apoptosis/ Necrosis Detection Kit (blue, green, red) is great to simultaneously monitor apoptotic, necrotic and healthy cells. It uses the dye 3-AAD (Ex/Em = 546/647 nm) to label the nucleus, suggesting the loss of plasma membrane integrity and late-stage apoptosis and necrosis. It also uses Apopxin Green Indicator (Ex/Em = 490/525 nm) as PS sensor upon binding to membrane PS. And CytoCalcein Violet 450 (Ex/Em = 405/450 nm), for labeling living cell cytoplasm.I used it to work with a pool of mouse cells, studying their cellular physiology in response to treatment. This kit works well to tell the different cell fate.

Experimental Design and Results Summary

Application

Mouse cell fate study

Starting Material

A pool of mouse cell suspensions

Tips

I use it for suspensions, imaging working with adherent cells will be hard.

Results Summary

I used this kit to study a pool of mouse cells and their physiology before/after treatment. Using FACS, I can see the percentage of cells that are healthy, apoptotic or necrotic. If wanted, we can also sort out these specific cell types, and look for their specificity, such as cell type, and protein expression pattern.

Summary

The Good

Works with FACS and cell sorting.

The Bad

May not work with adherent cells.

The Bottom Line

This kit works great with FACS based cell fate study, including healthy, apoptotic or necrotic cells. It works well with suspension cell pools, even different cell types mixing together is OK.
