Transformation of large sized plasmid constructs and BACs require electro-transformation. Using suitable apparatus which delvers controlled electric pulses is critical for successful transformations. For this purpose, I used Bio-Rad laboratories Xcell Gene Pulser to transform plasmid constructs of sizes ranging from 25 to 40 kb and succeeded in getting transformants.
Transformations and cloning
Use high efficiency electrocompetent cells. Make sure that plasmid DNA were free of any salt contamination. Pre-chill electroporation cuvettes.

Electric voltage, capacitance and resistance parameters are operator controllable. Bio-Rad cuvettes are arc proof compared to other providers.
