Adherant cell lines attached to tissue culture flasks as they secrete extracellular matrix for attachment. In order to detach cells, you need to digest the matrix using trypsin as a protease. This ready made solution is added to cells to detach them from the plastic.

Need to wash away excess media with PBS before adding trypsin solution. Incubate at 37 deg until detached. Quench reaction with full media containing serum.
