To study the role of heat shock protein during fungal toxin induced oxidative stress, HSP 70 protein was detected in control and toxin exposed tissue samples using this antibody. Briefly tissue lysate was prepared in tissue lysis buffer, maintaining cold chain during the process. Protein concentration in tissue lysate was quantified and 50 µg of protein was resolved on 10% SDS-PAGE, transferred to nitro-cellulose membrane, blocked and probed with anti-HSP 70 primary antibody overnight. Again after multiple washing of the membrane, probed with anti-rabbit HRP conjugated secondary antibody for 1 h. HSP 70 protein expression levels detected on X-ray flim with chemiluminescent detection reagent.
