PCR can be time-consuming, due to the multiple runs required to optimize reaction conditions for amplification of a single target. The new Takara PCR Thermal Cycler Dice provides a gradient feature. The gradient can span up to 20ºC, with each row in the sample block at a unique temperature within the desired range. This feature can be used at any step in the program. Since the gradient allows for multiple temperatures to be tested, reaction conditions can be optimized in a single experiment. This feature helps us in the standardization process of newly designed primers; we can use this feature by applying 12 different temperatures across the rows in a single run.

The Takara Gradient thermal cycler is built around a single universal block that accommodates 96-well plates or up to 96 0.2 ml PCR tubes. The thermal cycler allows for easy sample preparation without mineral oil and uses Peltier technology to provide uniform, quick heating and cooling. The feature that differentiates this instrument from its competitors is the gradient range (up to 20ºC). However, the instrument has numerous other great features. The sliding cover with a lever-type handle makes sample placement into the block easy. The adjustable control panel makes the instrument very user friendly; it is easy to operate and every setting is clearly indicated on the LED screen for convenient operation. The cycling program is displayed as a simple representative graph showing the temperature and time at each point during the cycle. It is easy to get started with 8 preinstalled PCR protocols. PCR pop-up menus and alpha-numeric entry via a 10-number keypad enable smooth and easy operation. The instrument also has an extension time and extension temperature functions which allow the extension time and temperature to be varied with each cycle of PCR. These functions are mainly suitable for long PCR and touchdown PCR, respectively. The remaining parameters like the small footprint (260 mm × 345 mm × 260 mm), light weight (11.5 kg) and temperature accuracy of +/- 0.5ºC are also pluses. When running, the thermal cycler is rather quiet and does not negatively impact the noise level in the lab. It can also be attached to your personal computer in order to store as many runs as needed. Automatic restart of the program in the case of a power failure is another feature that I find very useful.

The temperature ramping rate in this cycler is adequate, although not exceptional. We use the Takara PCR Thermal Cycler Dice mainly for diagnostic purposes, as well as for some contract based research. This instrument has proven to be a reliable tool for diagnosis. A possible drawback of this instrument is that the block cannot be changed. In summary, I can say the Takara Thermal Cycler Dice is an excellent thermal cycler with outstanding performance and consistent results.

The Good

This instrument is very easy to use. The temperature gradient (up to 20ºC across the rows of a sample block) is very useful for optimizing PCR conditions.

No interchangeable blocks, average ramping rates.
