We have used this LDH kit to measure the plasma membrane damage and its integrity loss in vitro. Assay was performed according to the manufacturer instructions. Briefly, 50 µl of cell culture supernatant from toxin treated cells was mixed with twice (100 µl) of reaction mixture prepared as per kit instructions and incubated at room temperature in dark for about 30 minutes and absorbance was recorded at 490 nm using microtiter plate reader.

Reconstitute LDH assay cofactor in 25 ml of tissue culture grade water. Aliquot around 1 ml in each eppendroff and store at -20 degree C. Avoid the freeze thaw of the reconstituted LDH assay cofactor

Results Summary

Cell supernatant from cells with toxin exposure showed higher absorbance at 490 nm compared to control cells indicating cell membrane damage.
