To check the cytotoxicity potential of fungal toxin on human monocytes, we have used this Thiazolyl Blue Tetrazolium Bromide (MTT) dye. When cultured cells incubated with yellow color MTT dye, mitochondrial succinate dehydrogensae enzyme converts MTT into purple color formazn crystals indicating cell viability. Briefly before the MTT assay, human keratinocytes were cultured in 96 well plates and treated with the different concentration of the toxin in triplicates. After completion of toxin incubation, cells were incubated with 100 µl of 1 mg/ml freshly prepared MTT solution for 4 h at 37°C in dark and formazan crystals formed were solubilized in DMSO and absorbance was measured at 570 nm in ELISA reader. To calculate the cell viability, absorbance for control cells were noted as 100% viable cells.
