To study the apoptosis pathway during toxicity of fungal toxin, we have checked the expression levels of caspase-9 in Hela cell line. After completion of toxin exposure for 24 h cells were lysed in RIPA buffer. Total cell protein was estimated using protein assay kit and equal amount of protein was resolved on 10 % SDS-PAGE. Resolved protein was transferred to nitro-cellulose membrane, blocked with 5% skimmed milk in PBST buffer, probe with primary caspase-9 antibody overnight and with HRP conjugated secondary antibody for 1 h. Caspase-9 levels were detected on X-ray flim using chemiluminescent sbstrate.
