Cell Signaling Technology

Product Name:

Phospho-FoxO1 (Ser256) Antibody

Catalog Number:

9461

Clone Number:

Our lab studies the role of mTOR signaling in T cell such as mTORC2 signaling during T cell differentiation and function. With immunoblot analysis, we assessed mTORC2 activity using pFoxo1 (S256) as a readout in WT T cells upon T cell activation.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Murine CD8+ T cells

Primary Incubation

1:1000 in %4 BSA/TBST 4 deg overnight shaking

Blocking Agent

5% milk/TBST room temp shaking

Secondary Incubation

1:5000 anti-Rabbit HRP (CST) in 5%milk/TBST room temp 1 hr shaking

Tertiary Incubation

None

Detection

UVP Imaging System

Results Summary

We are able to detect differential p-Foxo1 activity in our conditions. The signal is quite okay since exposure time was low but this is based on our conditions and samples.
