The expressions of the naive memory marker (using a combination of antibodies, usually CD45RA and CD45RO) on peripheral blood T cells is important in any immunodeficiency panel. In our lab, we have established an assay for T cell identification, but needed a good CD45RO antibody because after antigen experience, central and effector memory T cells gain expression of CD45RO and lose expression of CD45RA. We went for this antibody because it fits our panel, and it is ASR.

Using an LSR-II flow cytometer (equipped with a red laser)
The results are quite impressive as we got very nice staining even on the first run, without any optimization or titration. We used the volume recommended by the manufacturer, and it shows a spectrum of positivity on lymphocytes (blue), but of course it is also positive on monocytes and granulocytes (grey). The plot on the left is the negative control.

A good antibody for those who don't want to spend ages optimizing their results. It is also ASR.
