I highly recommend Bio-Rad's C1000 thermal cycler. I bought mine almost 10 years ago, and it was a demo model, so I got a great discount. I love it so much, I have since purchased the CFX96 Touch, which is their RT-PCR detection system.

mRNA is isolated using Ribozol. cDNA reverse transcribed from mRNA using cDNA synthesis kit. SYBR Green Supermix used along with primers to amplify target sequences. Gene expression determined using a comparative Ct method with GAPDH as an endogenous control.
Good primer design and pipetting skills are essential
