I need to emphasize that most of these types of kits from Sigma, including formate, acetate, acetyl-CoA, lactate... All their descriptions on the website said sufficient for 100 colorimetric tests. It is correct only if you are willing to risk to use no background controls! However, critical research requires controls, so this kit can only measure half the samples for real. And, you need to use 12 wells for standard cure, thus, you can only measure about 40 samples.

Protocol Overview

Set up the Reaction Mixes according to the schemein Table shown. Note: NADPH present in samples can cause background in the assay. To remove the NADPH background, include a blank sample for each sample by omitting the Formate Enzyme Mix. The background control readings can then be subtracted from the sample readings. 2. Add 50 μL of the appropriate Reaction Mix to each of the standard, sample, and blank control wells. Mix well using a horizontal shaker or by pipetting, and incubate at 37°C for 60 minutes, protected from light. 3. Measure the absorbance at 450 nm (A450).
