Product Name:

Phospho-FoxO1 (Thr24)/FoxO3a (Thr32) Antibody

Catalog Number:

9464

Our lab studies mTORC2 signaling in macrophage polarization and function using Rictor KO macrophages. FoxO1 is downstream of this pathway since it acts as a transcription factor. Phosphorylation of FoxO1 leads to translocation from the nucleus to the cytoplasm. This antibody will detect the critical residues that leads to movement from the nucleus to the cytoplasm. This is a reliable antibody that is widely used in the scientific literature.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Murine macrophages

Primary Incubation

1:2000 in 4% BSA/TBST

Blocking Agent

5% dry milk/TBST

Secondary Incubation

1:5000 Rabbit IgG HRP in 5% milk/TBST

Tertiary Incubation

None

Detection

Invitrogen ECL Pico

Results Summary

We are able to detect differential p-FoxO1/3 activity in our experimental conditions. The signal is quite robust since exposure time was low but this is based on our conditions and samples.

Summary

The Good

Reliable antibody to detect pFoxo1.3 at a good dilution

The Bad

None really

The Bottom Line

One of the few antibodies on the market to truly detect p-FoxO1/3 activity. Widely used in the scientific community.
