We routinely manually count cells using a hemacytometer. Trypan blue helps us determine the cell concentration.

Protocol Overview

We mix our biological sample of cells with trypan blue to help count viable cells. We typically dilute the trypan blue to .04% in PBS since 0.4% seems too strong for our counting.

Tips

Dilute 1:10 in PBS to make it easier to count while saving reagent.

Results Summary

We are able to get appropriate cell concentration by counting viable cells (not blue) from non-viable (blue) using the hemacytometer.
