Our lab studies the role of mTOR signaling in T cell differentiation and function. We assess mTOR signaling through western blot analysis in T cells upon different stimuli and conditions; therefore, we perform a lot of western blots. We use the Invitrogen SureLock system to run SDS-PAGE, but we use this blot module to perform our transfer of protein onto membranes.

Protocol Overview

Run normal SDS-PAGE gel. Open the precast gel. Prepare transfer solution composed of the transfer buffer and certain percentage of methanol and top with water. Remove gel and push against membrane along with filters. Place in the module and place enough sponges to ensure tight seal. Fill chamber with extra buffer to run.

Tips

Make sure enough buffer is within and there is a tight seal to ensure efficient transfer of protein.
