My experiment was focused on evaluating the effect of a variant on the localisation of a secreted protein. My hypothesis was that the variant could impair the physiologic protein secretion. I used the anti-TGN38 antibody as a marker of Golgi apparatus, to localise the wild-type and mutant protein along the secretory pathway. This antibody was used for immunofluorescence on HEK293 cells fixed with paraformaldehyde 4%.
