This antibody gave us reproducible and high quality staining of neuronal nuclei in vibratome-sliced fixed mouse brains. This enabled us to localize microglia and neurons simultaneously and study their spatial relation.

Experimental Design and Results Summary

Applications

Immunofluorescence

Sample

Vibratome-sliced fixed mouse brain

Primary Incubation

1:1000, overnight at 4degrees C

Blocking Agent

5% Normal Goat Serum and 5% BSA

Secondary Incubation

1:1000 Gt anti-Rb Cy3, 1h at RT

Tertiary Incubation

N/A

Detection

Confocal laser scanning microscopy, 60x objective

Results Summary

This Ab was used with a red secondary Ab on brain slices from mice expressing GFP in microglia. This enabled us to localize microglia and neurons simultaneously and study their spatial relation. Since the Cy3 signal faded similar to gfp with deeper confocal scanning, the Ab successfully penetrated the entirety of our thick (100um) slice preparation.

Summary

The Good

Great neuronal nuclei marker

The Bad

Doesn't get the full morphology of the neuron, obviously

The Bottom Line

A no-brainer, widely used monoclonal Ab for identifying neuronal nuclei
