Thermal cyclers are used to run polymerase chain reaction (PCR), in particular RT-PCR, to determine the transcription level of a gene of interest. The C1000 Thermal Cyclers from Bio-Rad are compact instruments compatible with different 1000-series reaction modules. Our machine is equipped with a "Dual-48" module that has 2 independent 48-well blocks. It also has a touch screen, which is user-friendly and convenient.

RNA from Human tumor cells

Protocol Overview

Human tumor cells are harvested and RNA is immediately isolated from the cell pellet with TRIzol reagent. Three microgram of purified RNA are subjected to cDNA synthesis using Takara/Clontech "RNA to cDNA EcoDry" kit. The samples volumes are between 30 and 40 microliters. PCR protocol includes: initial denaturation and activation step (94oC for 3 min), followed by 35 cycles of: denaturation at 94oC 30 sec, annealing of primers at 55oC for 30 sec, synthesis of the complementary strand at 72oC for 1 min.

Tips

The cycler should have sufficient air flow to prevent overheating. The blocks should be cleaned regularly.
