The aim was to determine whether there was a difference in IL-1b protein levels in the brains of mice using a mouse model that mimics Multiple Sclerosis symptoms. This kit was chosen because it was recommended by a colleague.

The manufacturers protocol was followed with minor changes . Briefly, the capture antibody was added to the wells and left overnight, wash with was buffer and then blocked with 1% BSA in PBS overnight. The samples and stds were then incubated overnight, before adding the detection antibody. After adding the Strep-HRP and peroxide substrate solution, the reaction was stopped with 0.86N H2SO4 after 10 mins. and read at 450 nm. I incubated the samples overnight at 4 degrees Celsius rather than the suggested 2 hours at room temperature and also blocked overnight rather then for 1 hour.
Lower concentrations of H2SO4 also produces identical results.
We were able to detect IL-1b in all of our samples
