The ThermoFisher Exosome Isolation Reagent can be used to isolate small extracellular vesicles from cell conditioned media. The process is quick and simple and does not require specialist equipment. Exosomes are precipitated out of solution and then pelleted by centrifugation.

Protocol Overview

Harvested cell culture media is centrifuged at 2,000xg to remove cells and debris. The exosome reagent is then added (0.5 volumes) and the sample is refrigerated overnight. Samples are then centrifuged at 10,000xg for 1 hour to precipitate the exosome pellet.

Tips

Cells must be cultured in exosome-free media to avoid contamination of cell conditioned media with exosomes derived from other sources

Results Summary

The reagent precipitates abundant small vesicles that can be characterized by electron microscopy, nanoparticle tracking analysis and Western Blot for common exosome markers.
