We are an immunology lab that studies the adaptive and innate system. We primarily are a flow based lab since most of our experiments rely on analyzing different cell populations in the context to infections or cancer. Most experiments involve first identifying different populations of cells but also analyzing expression of key activation and inhibitory molecules. Multi color flow cytometry allows us to conduct such research.

Murine T cells from spleen, tumor, lymph nodes or human PBMCs

Protocol Overview

After obtaining single cell suspensions of our samples, we FC blocked to remove background staining before we performed surface panel staining. If intracellular staining (ICS) is required, cells were fixed/permeabilized before ICS staining. Cells were thoroughly washed in between steps before acquisition on the analyzer.

Tips

Single Stain Controls are key to any flow experiment along with panel design based on marker expression

We use this instrument daily in the lab to perform our flow cytometry experiments.

We have a FACS flow supply unit that pumps sheath fluid at a constant rate to maintain pressure throughout.

Highly recommend this instrument for any lab that heavily relies on flow cytometry for their experiments. If user is LSR2 experienced, the transition is quite easy since the software and hardware is similar. Everyone in the lab is satisfied with its performance.
