We study the expression of a newly discovered receptor, transcobalamin receptor (TCblR), required for uptake of cobalamin by cells. We studied TCblR expression in adult mouse kidneys where we used hematoxylin qs as a counterstain.

Protocol Overview

After DAB staining, sections were washed in PBS and then incubated with hematoxylin qs for 5 minutes. The sections were then washed in tap water, twice, to remove excess hematoxylin, dehydrated in 70% to 100% ethanol, cleared in xylenes and mounted with permount.

Tips

Incubation needs to be at least 5 minutes for paraffin sections else the stain goes away when dehydrated in ethanol afterwards.

Results Summary

We found TCblR expression in glomerulus in kidneys cortex region.

It works really well on paraffin embedded sections and is very easy to use. It does not require any dilution and can be stored at room temperature.
