Beta actin is one of the most used loading (normalization) control in Western blotting experiments. Sigma's HRP conjugated beta-actin antibody makes the normalization step a quick one without the need for stripping the membrane.

Experimental Design and Results Summary

Applications

Western Blot

Sample

Human coronary artery endothelial cell lysates

Primary Incubation

1:40,000 dilution in 3% non-fat skim milk for 2 hours at RT.

Blocking Agent

3% non-fat skim milk for 30 min (PVDF), 1 hour (nitrocellulose)

Secondary Incubation

Not required

Tertiary Incubation

NA

Detection

Chemi luminescence

Results Summary

20 ug endothelial cell lysate was used in this experiment. After SDS-PAGE and blotting, the membrane was blocked with 3% non-fat skim milk and incubated with 1:40,000 diluted anti-beta actin-HRP antibody.

DOI or PMID #

NA

Additional Notes

If you are reusing the blot, no need of stripping and reprobing. Block the membrane again after first use and incubate with beta-actin HRP antibody.
