Our lab studies CD4+ T cells and often performs in vitro or ex vivo (re)activation using anti-CD3 + anti-CD28.

Experimental Design and Results Summary

Applications

In vitro activation of CD4+ T cells

Sample

Murine CD4+ T cells

Primary Incubation

Goat anti-hamster IgG coated plates

Blocking Agent

None

Secondary Incubation

Anti-CD3 + anti-CD28

Tertiary Incubation

None

Detection

ELISA for cytokines to assess T cell activation

Results Summary

We coat plates with goat anti-hamster IgG for anti-CD3 crosslinking. Cells are stimulated for 24 hours. Supernatants are collected and cytokines are measured by ELISA to assess T cell activation. We see robust activation of T cells using this method and these antibodies.
