Our lab recently performed southern blots on DNA from a newly generated knockout mouse to ensure deletion of the gene of interest. We used this chemiluminescent kit to detect the knockout allele.

Digest DNA, run gel, blot, incubate blot with probe, detect using kit as follows:-Block blot with blocking buffer (in kit) at 37*C for 15 min. Use 15-20ml-Wash 3 times for 5min in 1x washing buffer-Incubate membrane in substrate equilibration buffer for 5min-Incubate membrane in substrate working solution for 5 min-Wrap membrane in plastic and avoid bubbles and wrinkles-Capture image of blot by exposing under a camera or in expose to x-ray film
